چکیده

We report the simultaneous characterization of timeand force-dependent mechanical properties of adherent cells in the physiologically relevant regime of large forces. We used magnetic tweezers to apply forces to magnetic beads bound to the cytoskeleton, and recorded the resulting deformation (creep response). The creep response followed a weak power law at all force levels. Stress stiffening and fluidization occurred simultaneously and were quantified by the force-dependence of the creep compliance and the power law exponent. The amount of stiffening and fluidization in response to force was controlled solely by the contractile prestress of the cell and followed a simple relationship. This behavior may be of fundamental importance in biological processes that involve a mechanical interaction between cells and their environment.

Filamin A is essential for active cell stiffening but not passive stiffening under external force.

The material properties of a cell determine how mechanical forces are transmitted through and sensed by that cell. Some types of cells stiffen passively under large external forces, but they can also alter their own stiffness in response to the local mechanical environment or biochemical cues. Here we show that the actin-binding protein filamin A is essential for the active stiffening of cells ...

Effect of Subchronic Administration of Captopril on a1-Adrenoceptor Agonist-Induced Contraction of Isolated Aorta in Rat

Angiotensin II is a major endocrine hormone that affects directly both vascular smooth muscle and endothelial cells. Since vascular reactivity to angiotensin II changes in more physiological and pathophysiological conditions, the present study was performed to investigate the effect of intraperitoneal administration of angiotensin-converting enzyme inhibitor and captopril (30 and 50 mg kg-1, on...

Scaling laws for the response of nonlinear elastic media with implications for cell mechanics.

We show how strain stiffening affects the elastic response to internal forces, caused either by material defects and inhomogeneities or by active forces that molecular motors generate in living cells. For a spherical force dipole in a material with a strongly nonlinear strain energy density, strains change sign with distance, indicating that, even around a contractile inclusion or molecular mot...

Cellular adaptation to mechanical stress: role of integrins, Rho, cytoskeletal tension and mechanosensitive ion channels.

To understand how cells sense and adapt to mechanical stress, we applied tensional forces to magnetic microbeads bound to cell-surface integrin receptors and measured changes in bead displacement with sub-micrometer resolution using optical microscopy. Cells exhibited four types of mechanical responses: (1) an immediate viscoelastic response; (2) early adaptive behavior characterized by pulse-t...

A silicone-based stretchable micropost array membrane for monitoring live-cell subcellular cytoskeletal response.

External forces are increasingly recognized as major regulators of cellular structure and function, yet the underlying mechanism by which cells sense forces and transduce them into intracellular biochemical signals and behavioral responses ('mechanotransduction') is largely undetermined. To aid in the mechanistic study of mechanotransduction, herein we devised a cell stretching device that allo...
